Overview

OBESITYsym™ is a quantitative systems pharmacology (QSP) model developed by Simulations Plus, designed to help pharmaceutical researchers and drug developers predict both the efficacy and side effects of weight loss therapies. By leveraging a single QSP model alongside existing clinical data, OBESITYsym enables teams to optimize dosing strategies, anticipate patient variability, and address the most significant barrier to adherence in current obesity treatments: nausea.

As the newest generation of weight loss drugs demonstrates high clinical effectiveness, the nausea side effects associated with these therapies can undermine patient adherence and limit their public health impact. OBESITYsym was built to help drug developers navigate these challenges, supporting the development of treatments that can help reduce the burden of conditions linked to excess weight, including heart disease, Type 2 diabetes, osteoarthritis, and high blood pressure.

Key Predictive Capabilities

  • Predict drug efficacy for weight loss
  • Estimate the expected range of weight loss outcomes
  • Assess the probability of nausea side effects
  • Identify dosing strategies that maximize weight loss while minimizing nausea
  • Model the full range of patient variability in clinical responses
  • Determine which patients are likely to develop tolerance
  • Identify patients who should be monitored for down-titration for safety purposes

Platform Features and Technology

  • Built on Simulations Plus's QSP modeling platform, Thales, and extensively validated with clinical data
  • Includes SimPops, a feature that simulates the full range of clinical responses reported with GLP-1 treatments currently available to obese patients
  • Supports prediction of outcomes for both single and combination treatment strategies
  • Enables exploration of mechanisms and modes of action for novel treatments
  • Allows benchmarking of new compounds against established drugs on the market, including semaglutide, tirzepatide, and liraglutide
  • Accepts existing clinical experience and data as inputs to further optimize predictions

Working with QSP Experts

  • Access to a team of highly experienced QSP modelers who understand drug development program goals
  • Expert support available to help teams meet specific modeling and simulation objectives
  • Scientists with deep expertise in quantitative systems pharmacology are available for collaboration

OBESITYsym is delivered as software accessible by remote connection, making it available without local infrastructure requirements. It is designed for use by pharmaceutical and biotech organizations developing compounds targeting overweight or obese patient populations, and can be licensed or evaluated through Simulations Plus.
