LV Edge System
Lentiviral production with engineered cell lines and model-guided DNA design to achieve high titers for therapeutic transgenes.
Overview
The LV Edge System is a lentiviral vector production platform developed by Asimov that integrates engineered stable cell lines with software-guided DNA design tools. It is designed for biopharmaceutical and gene therapy developers seeking to achieve lentiviral titers of up to 1E9 TU/mL for therapeutic transgenes, including clinically relevant constructs such as anti-CD19 CARs. The system is intended to reduce lentivirus production costs while supporting scalable, reproducible manufacturing.
The platform consists of three core components: a clonal, suspension-adapted cell line with stably integrated lentiviral genes under inducible control; model-guided DNA design for optimizing expression of a Gene of Interest (GOI), powered by Kernel, Asimov's computer-aided design (CAD) software; and ready-to-transfer processes and protocols across multiple production scales. Users can choose between an off-the-shelf packaging system for in-house use or a fully-stable producer cell line development service performed by Asimov.
LV Edge Packaging System
- Optimized for speed of deployment
- Asimov ships a clonal suspension packaging host cell line to the user's facility
- Users receive access to Kernel software to design and request plasmid DNA for their GOI
- Lentiviral production is performed by the user at their own facility
- Supports transient packaging workflows
- Attributes include batch-to-batch consistency, low manufacturing complexity, low supply chain risk, scalability, rapid development timeline, and low cost
LV Edge Producer System
- Optimized for scale and reproducibility
- Asimov performs fully-stable cell line development and bioreactor process development at its own site
- A stable producer cell line containing the user's GOI is developed and shipped to the user for lentiviral production at their facility
- Supports fully-stable producer cell line workflows
- Attributes include batch-to-batch consistency, low manufacturing complexity, low supply chain risk, scalability, rapid development timeline, and low cost
Workflow Options
- In-house development path: Asimov ships software, cells, and genes; the user designs DNA using Kernel software with best-in-class parts and models, transfects cells, generates stable cell lines, and uses the software for data analysis
- Asimov-led development path: The user requests a producer cell line for a biologic or gene therapy; Asimov conducts cell line development and process development on-site, shares progress and data via Kernel software, and ships clonal cell banks with process transfer to the user's site
System Performance
- Demonstrated production of lentivirus exceeding 1E8 TU/mL for clinically relevant CAR constructs
- Performance assessed across transient, packaging, and producer cell line formats using an anti-CD19 CAR as a model transgene
- Target titer of 1E9 TU/mL for therapeutic transgenes
The LV Edge System is offered by Asimov, based in Boston, MA. The platform combines cell line engineering, software-guided construct design through the Kernel CAD tool, and process development services, with the option for full technology transfer to the user's manufacturing site.
